If there is one or more holes in the piston, a significant amount of fresh air/exhaust gases should enter the crankcase through these holes.
When opening the oil filler cap, you should definitely feel or hear something – it should release with some pressure.
It would be at least one way to find out more about the pistons without having to disassemble everything.
When you remove the fuel lines from the injectors and the fuel rail, the valve covers can be easily and quickly removed. This allows you to relatively easily rule out mechanical problems with the valve actuation system. However, this doesn't mean you can see whether the valves are sealing properly.
