I'm not sure if it absolutely needs to be done after 100,000+ km, but I remember that in my A6 with 380,000 km, the transmission sometimes felt like it didn't want to engage first, second, or third gear; the shifter would simply feel a certain resistance, regardless of whether it was cold or warm.
