The screw centers the disc in its seat and transmits no forces during driving. I would still replace it, just imagine having to change a flat tire on a dark country road - possibly even in the rain - and not being able to find the holes... Or if you can't position the remaining screw stub over the hole, the disc will rub endlessly.
Try drilling a hole in the screw. Make sure to center the drill bit properly beforehand, and then use a screw extractor to remove it, as Bertil already mentioned. The screw is pre-coated with a sealant from the factory, which is why it's so difficult to remove. Original replacement screws already have this micro-encapsulated coating applied.
"In my opinion, the best way to remove these screws is with an impact driver, but make sure to verschandeln the crosshead slot thoroughly beforehand. Back in my early days of using power tools, around 1985, I even stripped a few of them myself, which was quite a mess. I didn't have an extractor at the time, so I carefully drilled out the remaining threads completely." Gruß
